PREPARASI DAN UJI AKTIVITAS ANTINEFROLITIASIS NANOSUSPENSI FRAKSI ETIL ASETAT DAUN KERSEN (Muntingia calabura L.) TERHADAP TIKUS PUTIH JANTAN TERINDUKSI ETILEN GLIKOL
Kidney stones (nephrolithiasis) were a common health problem in Indonesia. The kersen leaf (Muntingia calabura L.) contained flavonoid compounds that acted as diuretics. The use of nanosuspension preparations was chosen to enhance the effectiveness and bioavailability of the fraction in delivering the active substance to the target. The aim of this study was to determine the %EE and characterization of ethyl acetate fraction of kersen leaf nanosuspension (NSFEADK) in reducing the levels of calcium, urea, and serum creatinine in rats based on kidney histopathology as an antinephrolithiasis agent. The method used in this study was the preparation of nanosuspension using an ionic gelation system and antinephrolithiasis test on experimental animals. The experimental animals were divided into 4 groups, namely the normal group (0.5% Na-CMC), the positive group (BatuginĀ® Elixir 791 mg/kgBW), the negative group (0.75% ethylene glycol and 1% amonium chloride), and the test group (ethyl acetate fraction of kersen leaf nanosuspension with a dose of 223.16 mg/kgBW). The best formula was obtained from formula 1, which was NSFEADK with %EE, particle size, PDI, and zeta potential of 96.72%, 176.50 nm, 0.29, and -29.08 mV, respectively, which could reduce the levels of kidney calcium, creatinine, and urea in the blood serum of the induced experimental animals. The renal calcium levels, blood serum creatinine and urea in the test group animals decreased by 3.50 mg/g, 0.56 mg/L, and 34.09 mg/L, respectively. The macroscopic and microscopic results of the kidney organs showed improved organ shape characterized by a decrease in the number of necrotic cells, glomerular atrophy, and crystal deposition. This indicates that NSFEADK, as an antinephrolithiasis, provided effectiveness that was not significantly different from the positive group (p>0.05) with a final dose of 215.84 mg/kgBW.
