Jatropha curcas linn leaves contain secondary metabolite compounds such as alkaloids, flavonoids (kaemperol), saponins, steroids, tannins, quinones, and phenols. Empirically, this study aims to determine the characteristics and phytochemical screening of ethanol extracts and fractions of Jatropha curcas leaves, the effect of administering ethanol extracts and fractions on MDA levels, catalase enzyme activity, macroscopic images, and histopathology of mice. This study was divided into 6 treatment groups, namely positive control (Vitamin C), negative control (Na CMC 1%), and test groups (ethanol extract, n-hexane fraction, ethyl acetate fraction, and ethanol-water fraction) at a dose of 100 mg/kgBW. The results showed that the characterization of the ethanol extract met the requirements of the Indonesian Ministry of Health 2008. The results of the phytochemical screening of the ethanol extract were positive for alkaloids, flavonoids, tannins, saponins, phenolics, steroids and triterpenoids. The average MDA levels in the positive, negative, ethanol extract, n-hexane fraction, ethyl acetate fraction, and ethanol-water fraction groups were 0.797; 2.438; 1.113; 1.011; 0.838; 1.718 nmol/mL, respectively. The average catalase enzyme activity values in the positive, negative, ethanol extract, n-hexane fraction, ethyl acetate fraction, and ethanol-water fraction groups were 28.999; 10.130; 22.300; 22.776; 26.415; 15.492 U/mL, respectively. The results of macroscopic and histopathological images of the liver showed differences after administration of ethanol extract and fractions compared to the negative group. The ethyl acetate fraction has the strongest antioxidant activity compared to other treatment groups, as shown by the results of Duncan's statistical analysis, which showed no significant difference (p>0.050).
