PENGARUH NATRIUM HIPOKLORIT DALAM MENEKAN TINGKAT KONTAMINASI EKSPLAN LAMINA DUKU PADA KULTUR IN VITRO
Duku (Lansium domesticum Corr.) has a fairly high consumption rate but is slow in its propagation process. This problem can be overcome through in-vitro propagation. The aim of this study was to evaluate several sterilization procedures and to obtain the best sterilization method on lamina explants of duku plants in invitro propagation. This research was carried out at the Tissue Culture Laboratory, Faculty of Agriculture, UNSRI located on 2°59'23.4"S 104°43'53.4"E from Juny to September 2024. This sterilization treatment use a combination of 5 types of sterilant materials, namely liquid detergent, streptomycin sulfate, Benomyl, NaOCl and 70% alcohol. The presentation of data in the form of tabulations and figures was carried out by calculating the percentage of explants contaminated with bacteria and fungi, living explants, browning explants. The results of this study showed that the combination of sterilant in treatment P3, which included liquid detergent for 3 minutes, 0.2% streptomycin sulfate for 10 minutes, 1% NaOCl for 15 minutes and 70% alcohol for 5 minutes, was the best treatment because it could maintain 63% of living explants. Explants from P3 treatment did not experience browning with a percentage of 0% and was able to suppress bacterial contamination by 34% and fungal contamination by 3%.
