Spermtozoa cryopreservation is a technique for storing spermtozoa in a frozen state, the success of the cryopreservation process is influenced by the use of extenders. This research aimed to determine the best dose of honey supplementation in lactate ringer solution as an extender in the cryopreservation of Indonesian leaf fish spermtozoa. Cryopreservation research was carried out at the Laboratory Balai Pembibitan dan Hijauan Pakan Ternak (BPHPT), Sembawa. The production of cryomedia solution and rearing of Indonesian leaf fish broodstock were carried out at the Aquaculture Laboratory and Experimental Ponds, Aquaculture Study Program, Department of Fisheries, Faculty of Agriculture, Universitas Sriwijaya. This research was carried out in July-August 2024. This research used a completly randomized design consisting of four treatments and three replicates. The treatment (P) given was P0: 100% lactate ringer solution (control), P1: 0.25% honey + 99.75% lactate ringer solution, P2: 0.5% honey + 99.50% lactate ringer solution, P3: 0.75% honey + 99.25% lactate ringer solution. The results showed that the semen of the Indonesian leaf fish before cryopreservation had a pH value of 7, milky white color, medium viscosity, spermtozoa concentration of 2.18x109 cells mL-1, spermtozoa mass movement with a score of 5 and spermtozoa viability of 89.89%. The results showed that the best treatment, namely at P3 with a dose of 0.75% honey + 99.25% ringer lactate as an extender, was able to maintain the quality of spermtozoa of Indonesian leaf fish with a spermtozoa mass movement value with a score of 4 and spermtozoa viability of 79.75% within 14 days of storage.
