PENGARUH NATRIUM HIPOKLORIT (NaOCl) TERHADAP TINGKAT KONTAMINASI EKSPLAN HELAIAN DAUN BUAH DURIAN (Durio zibethinus) SECARA KULTUR IN-VITRO
This study was aimed to evaluate the effect of sodium hypochlorite for sterilizing young leaf explants of durian plants (Durio zibethinus) in in-vitro culture. This research was carried out at the Tissue Culture Laboratory, Faculty of Agriculture, Sriwijaya University, Bukit Lama, Kec. Ilir Barat I, Palembang City, South Sumatra with location coordinates 2 59'23.4"S; 104 43'53.4"E from June to September 2024. This sterilization treatment procedure used 5 combinations of sterilant materials, each consisting of 10 culture bottles with 3 replicates resulted in total of 150 experimental bottles. The treatments were the combinations of liquid detergent, bactericide, fungicide, NaOCl, and 70% alcohol with different soaking durations. The design used was parametric descriptive analysis by calculating the percentage of explants contaminated with bacteria and fungi, the percentage of media contaminated with bacteria and fungi, and the percentage of live explants. It was resulted that sterilization of leaf explants (lamina) of durian plants using the P5 treatment procedure (Liquid Detergent for 3' + Streptomycin Sulfate 0.2% for 10' + Benomyl 0.2% for 10' + NaOCl 1.5% for 5 ' + Alcohol 70% for 5') showed the most significant results with 53% of live explants percentase and only 23% of contamination in both explants and media.
