UJI AKTIVITAS ANTIOKSIDAN EKSTRAK ETANOL DAN FRAKSI DAUN MAHONI (Swietenia macrophylla King) PADA TIKUS JANTAN GALUR WISTAR YANG DIINDUKSI CCl4
Mahogany leaves contain flavonoid, phenolic, triterpenoid, steroid, tannin and saponin compounds as antioxidants. This study aims to determine the characteristics, phytochemical screening, the effect of giving ethanol extracts and fractions of mahogany leaves on antioxidant activity with parameters of MDA levels, catalase enzyme activity, macroscopic and histopathological images of rat liver induced by CCl4. This study was divided into 6 groups, namely positive control (Vitamin C 10 mg/kgBW), negative control (Na-CMC 1%), and test groups (ethanol extract, n-hexane fraction, ethyl acetate fraction, and ethanol-water fraction) with a dose of 100 mg/kgBW. The characterization results of mahogany leaf ethanol extract have met the requirements of the Indonesian Ministry of Health 2017. The average MDA levels of the positive control group, negative control, ethanol extract, n-hexane fraction, ethyl acetate fraction, and ethanol-water fraction were 0,734; 2,428; 1,296; 0,795; 0,998; and 1,635 nmol/mL, respectively. The average results of catalase enzyme activity in the positive control group, negative control, ethanol extract, n-hexane fraction, ethyl acetate fraction, and ethanol-water fraction were 29,425; 10,514; 21,712; 28,487; 25,789; and 17,666 U/mL, respectively. Macroscopic and histopathological features of the liver showed improvement after administration of ethanol extract and fraction of mahogany leaves at a dose of 100 mg/kgBW. The n-hexane fraction of mahogany leaves has the strongest potential as an antioxidant. The results of Duncan's statistical analysis showed no significant difference (p>0.05) with the vitamin C positive control group.
