Excessive free radicals in the body can cause oxidative stress conditions which result in tissue damage. This condition can be prevented by consuming antioxidants. Cherry leaves (Muntigia calabura L.) often used as a medicinal plant and are believed to have antioxidant properties. Based on previous research, methanol extract of cherry leaves has strong antioxidant activity, while the ethyl acetate fraction in the medium category. In this research, the ethyl acetate fraction of cherry leaves was separated and purified to determine the compounds responsible for their antioxidant activity. This process were carried out using vacuum liquid chromatography (VLC) and gravity column chromatography (GCC). The antioxidant testing was carried out using the DPPH method (2,2, diphenyl-1-picrylhydrazyl) and compound identification using FTIR spectroscopy. The results showed that the antioxidant activity would be stronger if the fractions were separated. The IC50 value of column fraction C was 57.05 µg/mL; subfraction C6 24.075 µg/mL; and subfraction C6.2 17.182 µg/mL.This shows that the antioxidant compounds in the ethyl acetate fraction of cherry leaves are concentrated in one compound as the separation is carried out. The antioxidant activity (IC50) of cherry leaf secondary metabolite compounds is classified in the very strong category, equivalent to the antioxidant strength category of the positive control in the form of ascorbic acid. From the results of FTIR spectroscopy, it can be seen that secondary metabolites responsible for the antioxidant activity of cherry leaves come from the flavonoid compound group with amount 66,99 mg.
