Kersen leaves (Muntingia calabura L.) have been proven to have antioxidant properties, both in the form of crude extracts and fractions. The ethyl acetate fraction of kersen leaves separated by vacuum liquid column chromatography has strong antioxidant activity (IC50 43,40 µg/mL). This research is a continuation of the previous research by further separating the fraction of vacuum liquid column results from the ethyl acetate fraction of kersen leaves using gravity column chromatography. This study is useful to determine the antioxidant activity of the results of column fraction separation of kersen leaves and identify specific compounds to further determine the ability of antioxidants in inhibiting free radicals. Fraction separation was carried out by column chromatography on silica gel with eluent of increasing polarity for two separations to obtain more specific compounds. Compound monitoring used thin layer chromatography technique to determine the chromatogram pattern. Each column fraction was tested for antioxidant activity using the 1,1 diphenyl-2-picryl hydrazyl (DPPH) method and the active fraction of antioxidant compounds was identified using IR spectroscopy techniques. The results of the separation of the column fractions showed that the antioxidant activity of column fraction D increased, while column fraction E decreased in activity. The results of this test showed that the final column fraction in the advanced separation FD4 (2) had the highest level of antioxidant activity with an IC50 value of 17,363 µg/mL. This indicates the specific compound concentrated in fraction form has high antioxidant potential. IR spectroscopy identification results concluded that the secondary metabolite compounds that were successfully separated were flavonoids as much as 77,97 mg.
