Plants that contain high levels of flavonoid and phenol compounds have the potential to act as antioxidants and anti-inflammatories. One of them is the breadfruit plant (Artocarpus altilis). This study aims to determine the antioxidant and anti-inflammatory activity of breadfruit leaf fractions. The research began with extraction by maceration using solvents with varying polarities (n-hexane, ethyl acetate, and methanol). Each fraction was then tested for total flavonoid and phenolic content using the colorimetric method and antioxidant activity testing using the DPPH method. The fraction that showed the best antioxidant activity was then tested for anti-inflammatory activity in vivo by measuring the volume of edema from induction using carrageenan. Tests were carried out with doses of 200, 400, and 800 mg/kgBW and aspirin was used as a positive control. The research results showed that the methanol fraction had the highest phenolic and flavonoid compound content compared to the other fractions, with a total phenolic value of 83.33 mgGAE/g and flavonoids of 61.33 mgQE/g and antioxidant activity with an IC50 value of 29.85 μg/mL. Testing the anti-inflammatory activity of the methanol fraction of breadfruit leaves showed that the highest activity was produced at a dose of 800 mg/kgBW with a percent inflammation inhibition value of 68.05% while the positive control was 30.05%. Statistical analysis shows that there are real differences between the negative control group and all treatment groups. The positive control group also showed that there were real differences with all treatments. Based on the test results, it shows that the methanol fraction of breadfruit leaves has antioxidant activity and is also active anti-inflammatory and has the potential to be used as a raw material for anti-inflammatory drugs.
