Melinjo leaves (Gnetum gnemon L) is one plant that has many properties, one of which is as a diuretic. Active compounds such as alkaloids, flavonoids, steroids, and tannins can be found in melinjo. Antinephrolithiasis testing of ethanol fraction of melinjo leaves was carried out in vivo with the aim of determining the effect on ureal levels, creatinine and calcium, crystalluria and urine pH, as well as macroscopic and histopathological images of rat kidneys induced ethylene glycol and ammonium chloride. In this study, there were seven test groups: normal group, negative group, positive group (Batugin), melinjo leaf ethanol fraction group dose 55, 110,165, and 220 mg/kg body weight. Ethylene glycol and ammonium chloride were given to the aal treatment group for 14 days except normal group and then followed by giving batugin and ethanol fraction of melinjo leaves in the positive group and dose treatment group for 14 days. The total flavonoid content of ethanol fraction of melinjo leaves is 4.866%. The results after induction were calcium oxalate monohydrate (COM) crystals and calcium oxalate dihydrate (COD) crystals in rat urine. The results showed that the average levels of creatinine, ureum, and calcium in the dose groups of 55, 110, 165, and 220 mg / kg body weight decreased compared to the negative control group. Post hoc statistical analysis of LSD creatinine, ureum, and calcium levels showed that the dose group of 220 mg/kgBB was not significantly different from the positive control group (p>0.05). The results of macroscopic and histopathological images of the kidneys showed kidney improvement after administration of ethanol fraction of melinjo leaves compared to the negative control group. The ethanol fraction of melinjo leaves dose 220mg/kg body weight is more effective in lowering creatinine, ureum, and calcium levels. The ethanol fraction of melinjo leaves dose 220 mg/kg body weight has antinephrolithiasis activity.
