DNA BARCODING PADI SIPUTIH DAN SIAM VARIETAS LOKAL SUMATERA SELATAN BERDASARKAN SEKUEN GEN MATK
Local rice varieties are one of the germplasm in South Sumatra, such as Siputih rice and Siam rice. Local rice varieties are widely preferred for their cooking quality, flavor, and aromatic characteristics. Therefore, local rice varieties need to be conserved because they have the potential for crop improvement in the future. Conservation efforts should require a clear identity that can be done with a molecular approach, namely DNA Barcoding using the matK gene. The purpose of this study was to determine genetic variation and kinship relationships based on DNA Barcoding of the matK gene. The results of this study indicate that there are genetic variations in local rice varieties that occur mutations, in Siputih rice there are 6 points of substitution mutations including 3 transversion mutations and 3 transition mutations while Siam rice there is 1 point gap mutation deletion. The results of phylogenetic tree analysis show that Siputih rice has a close relationship with Oryza glaberrima, with a genetic distance of 0.0044 and similarity of 99.56% while Siam rice has a close relationship with Oryza nivara and Oryza rufipogon with a genetic distance of 0.0000 and similarity of 100% and between the two samples of Siputih rice and Siam rice have a kinship with a genetic distance of 0.0066 and similarity of 99.34%.
