PREPARASI DAN UJI AKTIVITAS ANTIDIABETES NANOSUSPENSI EKSTRAK DAUN MELINJO (GNETUM GNEMON L.) DENGAN VARIASI KONSENTRASI GOM ARAB
Melinjo leaves are known to have pharmacological activity as antidiabetics because they contain flavonoid compounds that can reduce blood glucose levels with their ability to act as antioxidants that protect β-cell damage as insulin producers and can increase insulin sensitivity. The effectiveness, bioavailability, and solubility of extracts tend to be low, so the formula in the form of nanosuspension is an option to be able to increase the solubility of drugs in water and bioavailability by increasing the surface area of drugs in contact with biological media. This study aims to determine the effect of arabic gum variation on the characterization of melinjo leaf extract nanosuspension and the reduction of blood glucose levels in type 2 diabetes mellitus rats. The methods used in this study are measurement of total flavonoid content, preparation of nanosuspension using ionic gelation system, calculation of %EE and antidiabetic test on test animals. Three formulas were used in the preparation with varying concentrations of gom arabic, namely 0.2%, 0.15%, and 0.1%. The best formula was obtained from Formula I, with an average particle size of 211.93 nm, a zeta potential of -16.54 mEv, a PDI of 0.3999, and a EE of 92.6214%. The best formula was given to test animals that had been induced by streptozotocin, and the results showed a decrease in glucose levels of 73.03%. The percent reduction in glucose levels of the nanosuspension preparation was better than the percent reduction in blood glucose levels of the group given the extract, which was 65.23%, and the percent reduction in blood glucose levels of the positive group given metformin, which was 71.93%. It is concluded that the variation of gom arabic in the nanosuspension preparation formula of melinjo leaf extract affects the percent of sorption efficiency and nanosuspension characteristics and shows a decrease in blood glucose levels that is not significantly different from the metformin group (p > 0.05).
