Open wounds are very susceptible to infection, can become ulcers and even chronic if they are not treated and cared for properly. Nanofiber as a modern wound dressing can deliver medicinal ingredients for caring wound. Betel leaves (Piper betle) and Binahong leaves (Anredera cordifolia) contain flavonoids, saponins, tannins which can accelerate the wound healing process. The aim of this study is to determine the activity of nanofiber combinations of Betel leaf extracts and Binahong leaf extracts for open wound healing on male white rats (Sprague Dawley). In this study, the treatments were divided into 6 groups, namely the positive control group (Wintuell®), the negative control group (no treatment), the placebo group, and for the group test which were given PVP/CA/EDS/EDB nanofiber consisted of K1 (once a day), K2 (twice a day), and K3 (three times a day). Observed parameters included area of wound, percentage of wound healing (% recovery), time of wound healing, and features of histopathology skin. Open wounds were made by using biopsy punch in diameter of ±10 mm. The observation of wound has been done for 21 days in every 3 days. The diameter of wound was measured by using imageJ®. Data was analyzed through the Shapiro-Wilk normality test and the one-way ANOVA test by using SPSS®. The results showed that the highest % recovery and the fastest wound healing time was K3 which reached 100% within 18 days. However, there was no significant difference (p>0.05) among treatment groups. Histopathological features on the day of 14th and 21st showed an increase in epithelial thickness. Providing of the test triggered the epithelialization process. K3 gave the thickest epithelialization (86.96 µm) was compared to the positive group (71.49 µm). It shows that K3 is the best treatment in wound care which can accelerate the healing process of open wounds.
