Cassava leaves contain rutin compounds which are flavonoid derivatives that have antibacterial properties. Transdermal patch preparation was chosen for the ethyl acetate fraction of cassava leaves because it can produce effective preparations to deliver active compounds through the skin to reach the systemic. This study aims to obtain the best formula with plasticizer concentration in transdermal patch preparation of cassava leaf ethyl acetate fraction as anti-acne by inhibiting the growth of Staphylococcus aureus bacteria. Transdermal patches were made by continuous mixing method(solvent casting) with 3 formulas namely FI, FII and FIII with propylenglycol variations of 20%, 15% and 10%. The patch produced weight uniformity of 33.766 mg, thickness of 0.132 mm, percent elongation of 77.421%, swelling index of 5.598%, humidity of 7.064%. The results of the content uniformity test were 100%. The stability test results were good with a level at the 6th cycle of 99.99%. The best formulation was obtained at a propylenglycol concentration of 20% in FI which produced patches with a smooth, highly elastic, and transparent surface. The results of transdermal antibacterial activity of cassava leaf ethyl acetate fraction patches were significantly different from clindamycin (p>0.05) with an average inhibition zone of 14.7 ± 0.45 mm and classified as strong. It can be concluded that the transdermal patch of cassava leaf ethyl acetate fraction has good characteristics, stability, and antibacterial activity in formula I with 20% propylenglycol concentration.
