Free radicals trigger oxidative stress through an increase in Reactive Oxygen Species (ROS) and Reactive Nitrogen Species (RNS) which leads to degenerative diseases such as cancer, cardiovascular diseases, and others. Antioxidants are needed to protect from free radicals. One of the plants that has potential as a natural antioxidant is pelawan leaves (Tristaniopsis merguensis Griff). Secondary metabolite compounds contained in pelawan leaves include flavonoids, phenolics, tannins, saponins, and steroids/triterpenoids. This study aims to determine the characteristics of ethanol extracts, the effect of giving ethanol extracts and fractions of pelawan leaves on reducing Plasme MDA levels and increasing liver catalase enzymes, as well as macroscopic and histopathological of the liver of male wistar rats induced by CCl4. The study was divided into 6 groups treatment, namely the positive control group (vitamin C 10mg/kgBW), negative control (NaCMC 1%), and 4 treatment groups (ethanol extract, n-hexane fraction, ethyl acetate fraction, and ethanol-water fraction) doses of 100mg/kgBW. The results showed that ethanol extracts and fractions of pelawan leaves have met the standards of characterization examination by the Indonesian Ministry of Health 2017. Plasma MDA levels in the positive control, negative control, n-hexane fraction, ethyl acetate fraction, and ethanol-water fraction respectively were 0.734; 0.752; 0.946; 1.073; 1.582; 2.428 nmol/mL. Liver catalase enzyme activity in the positive control, negative control, n-hexane fraction, ethyl acetate fraction, and ethanol-water fraction respectively were 29.425; 28.393; 24.252; 21.262; 18.737; 10.514 U/mL. The ethyl acetate fraction obtained the best results seen from the statistical analysis that equivalent to the positive control group (p > 0.05) so it can be concluded that ethyl acetate fraction of pelawan leaves at a dose of 100 mg/kgBW can reduce plasma MDA levels and increase liver catalase enzyme activity as well as provide good macroscopic and histopathological features of liver.
