UJI AKTIVITAS ANTIOKSIDAN EKSTRAK ETANOL DAN FRAKSI DAUN SEBURO (GARCINIA FORBESII KING) PADA TIKUS JANTAN GALUR WISTAR YANG DIINDUKSI CCL4
Reactive radicals cause oxidative stress, leading to cell damage. Antioxidants can convert radicals into less reactive ones. Seburo leaf (Garcinia forbesii King) has the potential as a natural antioxidant containing flavonoids, phenolic compounds, saponins, tannins, triterpenoids and steroids. This study aims to determine the characteristics of ethanol extracts, the effect of giving ethanol extracts and fractions of seburo leaves on antioxidant activity by measuring MDA levels, determining catalase enzyme activity, macroscopic and histopathological images of rat liver induced CCl4. The treatment consisted of 6 groups including positive control group (Vitamin C 10 mg/kgBB), negative control (Na-CMC 1%), and test group (ethanol extract, n-hexane fraction, ethyl acetate fraction and ethanol-water fraction) dose of 100 mg/kgBB. The results of the characterization of ethanol extract of seburo leaves meet the requirements of the Indonesian Ministry of Health 2008. The average MDA levels of positive control group, negative control, ethanol extract, n-hexane fraction, ethyl acetate fraction and ethanol-water fraction were 0.734; 2.428; 0.840; 0.748; 1.292 and 1.377 nmol/mL, respectively. The average catalase enzyme activity was 29.424; 10.519; 26.835; 27.694; 21.148 and 16.168 U/mL, respectively. Macroscopic and histopathological features of the liver showed an improvement in the administration of ethanol extracts and fractions of seburo leaves at a dose of 100 mg/kgBB compared to the negative control. N-hexan fraction of seburo leaves showed the highest antioxidant activity statistical analysis results (p>0.05) which means there is no significant difference with the positive control group given vitamin C 10 mg/kgBB.
