EFEK EKSTRAK DAUN KERSEN (Muntingia calabura L.) TERHADAP KADAR TNF-α PADA TIKUS WISTAR MODEL GOUT ARTRITIS AKUT
Background: Acute gout arthritis is an inflammation due to the accumulation of MSU crystal in the joint which leads to pain, swelling, and an increase in proinflammatory cytokines such as TNF-α. Cherry leaves contain flavonoids that have anti-inflammatory activity which plays a role in inhibiting the production of TNF-α. This study aims to determine the anti-infammatory effect of cherry leaves extract (Muntingia calabura L.) on serum TNF-α levels in wistar rats as a model of acute gout arthritis. Methods: An in vivo experimental study with a pre and post-test control group design has been conducted at the Faculty of Medicine, Universitas Sriwijaya in August-December 2023. After the acclimatization period, male wistar rats were divided into 5 treatment groups, each consisting of 6 rats; negative control (Na-CMC 0,5%), positive control (Colchicine), D1 (cherry leaves extract dose 200 mg/kgBW), D2 (cherry leaves extract dose 400 mg/kgBW), D3 (cherry leaves extract dose 800 mg/kgBW). Acute gout arthritis rat models were created by intra-articular injection of 50µl MSU crystal suspension into the synovial joint of the knee. After the induction of MSU crystal, TNF-α levels in the blood were examined twice, before and after treatment, using Rat ELISA TNF-α. Results: The cherry leaves extract showed effectiveness in significantly reducing TNF-α levels (Paired T-test p0,05) Conclusion: Cherry leaves extract can reduce inflammatory responses by reducing TNF-α levels serum in acute gout arthritis model rat models.
