EFEK PAPARAN SODIUM FLUORIDA TERHADAP PERILAKU TERKAIT FUNGSI KOGNITIF DAN RESEPTOR NMDA PADA MENCIT (MUS MUSCULUS)
Background: Fluoride is one of the minerals that are abundant in nature. WHO has declared the existence of fluorosis endemic areas due to excessive consumption. Fluoride has a neurotoxicity effect and impaired the cognitive function in memory domain. Memory formation is closely related to glutamate and its receptors (NMDA and AMPA). This study aimed to determine the effect of sodium fluoride exposure on cognitive function-related behavior and NMDA receptors. Method: This study was an invivo experimental study with a posttest only control group design. Thirty-two C57BL/6 strain mice aged 70 weeks were divided into four groups of Sodium Fluoride (NaF) doses: NaF group 1 (2.5 mg/L), NaF group 2 (5 mg/L), and NaF group 3 (10 mg/L), and one control group (NaF 0 mg/L). Mice were induced NaF for 8 weeks. After exposure, mice were tested for OLT and ORT to observe the ratio of discrimination on cognitive function-related behaviors. Then, mice were decapitated and hippocampus was analyzed to measure NMDAR subunit protein: Grin2a with western blot method. Results: Each NaF group had a lower OLT and ORT discrimination ratio compared to the control group (Bonferroni test: p0,05). The examination of Grin2a subunit proteins is still in progress, due to the unsuccessful optimization process yet. Conclusions: Exposure to fluoride at doses (2.5-10) has an effect on cognitive function decline.
