Protein is a macronutrient that is important for the growth and development of babies. Breast milk protein is needed to support the growth and development of babies in the early stages of life. Protein levels in breastfeeding mothers are dynamic and influenced by various factors, especially characteristics, nutritional status and protein intake. Objective: This study was to analyze the relationship between characteristics, nutritional status and protein intake with total breast milk protein levels in lactating mothers in Seluma Regency. Method: This research was conducted in March 2023 with a cross-sectional research design. The sample used was 77 lactating mothers in Seluma Regency who were selected by random sampling. Protein intake was obtained using a 3x24 hour food recall, and calculated using the Nutria Survey application. Total breast milk protein content was obtained by examining breast milk samples (10 μl) using the biuret method. The nutritional status of lactating mothers was based on underweight, normal and overweight categories. Characteristic data was obtained through interviews. Data were analyzed using univariate, bivariate and multivariate analysis using SPSS version 20. Results: As many as 67.5% had low total breast milk protein levels, the majority of breastfeeding mothers had less protein intake (76.6%) and 44.2% of lactating mothers had normal nutritional status (BMI 18.5-22.9 Kg/ m2). This research shows a significant relationship between income and total protein content of breast milk (p-value 0.026) and protein intake and total protein content of breast milk (p-value 0.007). Protein intake is the variable that has the most dominant influence on the total protein content of breast milk after being controlled by other variables (OR=5.549). Conclusion: Education regarding protein intake needs to be carried out. Adequate protein intake can produce quality breast milk which is needed to help the baby's growth and development.
