Fatigue can be a fatal risk that can threaten the safety of workers. An unfavorable work situation has the potential to trigger workers to feel tired, easily sick, stressed, and have difficulty concentrating, which can lead to decreased work productivity. Accidents experienced by truck drivers often occur, one of which is known to be due to fatigue. This study aims to determine the determinants of fatigue in truck drivers at PT. Petaling Mandraguna in 2022 by analyzing the external and internal factors of workers. This research method is quantitative with the type of observational analytic research and uses a cross-sectional approach. The sample took by using a quota sampling technique for 39 respondents. The data obtained were analyzed using the Statistical Program for Social Science (SPSS) with the chi-square test. The variables studied were fatigue with the dimensions of Work-Related Factors consisting of work duration, workload, and break time. Also, Non-Work Factors contained sleep disturbances, physical condition, and working period. The results of this study showed that 27 workers (69,2%) experienced fatigue, also there is a correlation between fatigue with work duration (P-value=0,022), workload (P-value=
