Background: Periodontitis is an oral cavity disease with a very high prevalence in Indonesia. Periodontitis can cause loose teeth and easy detachment. Robusta coffee extract Semendo contains active compounds as antiinflammatory and antibacterial agents that can reduce the number of PMN cells in chronic periodontitis treatment. Objective: To determine the effectiveness of robusta coffee extract Semendo in reducing the number of PMN cells in the treatment of chronic periodontitis in Wistar rats. Methods: The method used in this study was a post test only control group design. A total of 21 male Wistar rats were divided into 3 treatment groups. The induction of periodontitis in rats was done by injecting Porphyromonas gingivalis bacteria into the gingival sulcus of rats for 14 days. Periodontitis rats were treated with a gel according to the treatment group twice a day for 14 days. Robusta coffee extract Semendo gel was applied to periodontitis rats with variations in concentration, namely 40%, 50%, and placebo gel (negative control). All samples were euthanized on the-15th day after gel application, and histological preparations were made. The number of PMN cells was counted and analyzed statistically. Results: The group treated with 50% robusta coffee extract Semendo gel had the lowest PMN cell count.
