Kenikir leaves has pharmacological activity as an antidiabetic because it contains flavonoid compounds in the form of quersetin which can become the α-glukosidase enzyme inhibitor so that excess blood glucose level can be lowered. The effectiveness and bioavailability of extracts tend to be low so that the formula in the form of nanosuspension is an option for improving the delivery system which can increase the effectiveness of the preparation. This study aims to determine the effect of variations of sodium tripolyphosphate on the characterization of nanosuspension of kenikir leaf extract and on decreasing blood glucose levels in rats with type 2 diabetes mellitus. The nanosuspension preparation method used is ionic gelation because it is simple and produces more stable after preparation. Three formulas were used in the preparation of preparations with variations of sodium tripolyphosphate, namely 0.23%, 0.1% and 0.01%. The in vivo antidiabetic test by inducing a high fat and fructose diet in five groups of rats for 30 days to obtain diabetes. The negative control group was induced by nanosuspension basis, the positive control group was given metformin at a dose of 150 mg/kgBW, group I was given kenikir leaf extract 333 mg/kgBW and group II was given 333 mg/kgBW kenikir leaf extract nanosuspension. The first formula choose to be the best formula obtained an average particle size of 144.6 nm, a zeta potential of +15.32 mEv and a PDI of 0.487 which used for the antidiabetic activity examination. The decrease in blood glucose levels in the nanosuspension preparations of kenikir leaf extract was not significantly different from the positive group (metformin) (p>0,05) compared to the kenikir leaf extract which decreased not really significant.
