Breadfruit leaves (Artocarpus altilis (Park.) Fosberg) contain flavonoids which have activity and have the potential to act as photoprotective agents because of their ability to absorb UV light. This study aims to determine the effect of variations in the formulation of breadfruit leaf ethyl acetate extract on the effect of activity tests as a sunscreen with Carbomer 940 and Triethanolamine as a carrier and to test the activity of sunscreen gel in vitro and in vivo. In vitro testing was carried out by determining the SPF value of each sunscreen gel, while in vivo it was carried out using 15 mice divided into 5 groups, namely, negative control group (F0), positive control (F+) and treatment groups F1, F2, and F3 were given breadfruit leaf ethyl acetate extract with formulations of 2%, 4% and 6% respectively. Mice were given irradiation treatment using an exotera UV-B lamp for 72 hours to observe erythema and edema on the mice's skin. The results of the research show that ethyl acetate extract of breadfruit leaves can influence the SPF value where the SPF values obtained for the 2% Extract, F1, F2, and F3 were respectively 7.26; 9.30 ; 12.44 ; 14.82. In vivo testing showed that each variation of breadfruit leaf ethyl acetate extract was able to protect mouse skin against exposure to UV-B rays, even in the smallest formulation, namely F1 with an extract concentration of 2% with no erythema and edema appearing. Based on the research results, it can be concluded that there is an influence of variations in breadfruit leaf ethyl acetate extract which is able to protect mouse skin against exposure to UV-B rays even in the smallest formulation, namely F1 (2%) with no visible erythema and edema
