This study aims to identify Giant Soft-Shell Turtle (Pelochelys cantorii) from the waters of the Lematang River, South Sumatra Province, through the molecular characteristics of mitochondrial 16S rRNA genes. The research method is descriptive with sampling techniques based on events and the location is in Lematang Ilir Waters, Modong Village, Muara Enim Regency. Genetic analysis using a white blood cells sample taken in the posterior dorsalis aorta. DNA isolation and PCR were carried out in two laboratories of Bogor Agricultural University. DNA sequencing was performed at Singapore's 1st Base Laboratory. Phylogenetic position analysis was performed through comparison of other Trionychidae members. The results showed that the 16S rRNA gene segment produced 1,151 bp. The results of alignment with several members of the order Triyonichidae show the genetic distance of Pelochelys cantorii (P1) from the Lematang River with KT926834, JN016746, JN016747 of 0.0000. Based on the reconstruction of phylogenetic trees through the Neighbour-Joining method, the phylogenetic position of P. cantorii (P1) from the Lematang River is in one clade with the giant soft-shell turtle species Chitra indica. The implication of this research is to enrich the knowledge and molecular taxonomy of Indonesian giant soft-shell turtles and become material for enrichment of Vertebrate Zoology.
