IDENTIFIKASI BEBERAPA VARIETAS PADI (Oryza Sativa L.) LOKAL SUMATERA SELATAN BERDASARKAN METODE PEG 6000 DAN MARKA MOLEKULER (SSR)
This study aims to identify some local rice varieties of South Sumatra that are tolerant of drought stress using PEG 6000 solution and PCR. This research was conducted at the Laboratory of the Department of Agricultural Cultivation, Faculty of Agriculture, Sriwijaya University, located in Indralaya, Ogan Ilir Regency, South Sumatra. The research was held from July to December 2022. There were 2 factors in this study, the first factor was the concentration of PEG which was divided into 3 levels, namely distilled water solution as control (K), PEG concentration of 10% (L), and PEG concentration of 20% (M). The second factor was PCR analysis performed by combining PCR techniques using primers with random sequences for amplification purposes. Parameters observed were sprout length, shoot length, main root length, seminal root length, germination percentage, shoot dry weight, root dry weight, vigor index and PCR amplification results. The inpago 10 varieties and the local south Sumatra varieties dayang rindu have the character of being tolerant of drought stress when given PEG concentrations of 10% and 20% based on the variables that have been observed. From the results of electrophoresis used the comparison varieties inpago 5 and inpago 10, it shows that the dayang rindu varieties has the same bp from the various markers used with the genes of the comparison varieties which was resistant to drought stress.
