Background: Obesity is an accumulation of adipose tissue that is multifactorial in nature, and can cause pathological consequences. Intermittent fasting emerges as a regimen for treating obesity with various types, one of which is the time-restricted feeding type. This study aims to determine the effect of time-restricted feeding on the histological features of inguinal subcutaneous adipose tissue in obese mice. Methods: This research is an in vivo post-test only control group experimental study. Twenty male mice were divided into four groups: normal control, obesity control, time-restricted feeding with high-fat feed, and time-restricted feeding with standard feed. Prior to the treatment period, the obese control group and both time-restricted feeding groups were induced to become obese with a high-fat diet marked with a Lee index > 0.3 for two weeks. Treatment with time-restricted feeding was carried out for 14 days with fasting at 16.00-08.00 every day. Body weight was measured before and after the treatment period. The mice were terminated and the inguinal subcutaneous fat tissue was obtained and used as HE preparations. Histological examination was conducted by taking images through microscope and reading them with the ImageJ application to count the number of adipose cells in 10 random fields of view. Results: Respectively, the cell count of each group (cell count/field of view) were: 120,500 (49,700–136,200); 68,380 ± 9,194; 70,860 ± 11,029; and 79,360 ± 5,112. The weight of each group in grams (g) were: 28,200 ± 2,956 and 30,000 ± 4,450; 35 (28-35) and 35,000 ± 1,225; 35,000 ± 1,140 and 34,000 ± 0,837; and 32,200 ± 2,289 and 32,600 ± 2,400. There was no statistically significant difference between the cell counts among all groups. The weight of obese control group was increased significantly. Conclusion: Time-restricted feeding type of intermittent fasting has no effect on changes in the adipocyte cell count of inguinal subcutaneous adipose tissue and body weight in obese mice.
