Cellulase enzymes are a group of enzymes that break down cellulose by hydrolyzing β-1,4 glycosidic bonds into simpler forms such as glucose and cellobiose. This study aimed to confirm the cellulase-producing bacteria that have the highest cellulolytic activity and determine the optimal activity of pH, temperature, substrate concentration, and the influence of metal ions as cellulase enzyme activators. This research was carried out using a non-factorial Completely Randomized Design (CRD) method to optimize cellulase enzyme activity from cellulolytic bacteria. The parameter of this research was the activity of the cellulase enzyme activity as seen through the amount of reducing sugar released. Among the 20 cellulolytic bacterial isolates, the bacterial isolate with sample code D 10-6 (39) was the highest bacterial isolate with an activity of 10.2 U/mL. These bacterial isolates were used as samples in further studies to determine the optimum conditions of pH, temperature, substrate concentration, and metal ions as activators of cellulase enzyme activity. The optimum pH for cellulase enzymes was pH 6 to pH 8 with an activity of 23.894 ± 2.97 U/mL to 26.055 ± 2.33 U/mL. The optimum temperature for bacteria X was 30° to 35° with an activity of 42.94 ± 0.93 U/mL to 44.695 ± 0.57 U/mL. The optimum substrate concentration for bacteria X was 1% CMC concentration with an activity of 80.761 ± 2.41 U/mL. The metal ion FeCl2 was an activator of the cellulase enzyme with an activity of 106.27 ± 18.60 U/mL while other metal ions were inhibitors of the cellulase enzyme
