Barbonymus sp. is distributed in water bodies at a watershed on the island of Sumatra, Kalimantan and Java. The study aimed to identify the fish species Barbonymus originating from Ogan River and Gerong River, knowing COI sequences and genetic and phylogenetic distances by using COI gene barcoding DNA techniques. This research was conducted in December 2022-January 2023. Ten samples were obtained from the Ogan River and Gerong River in South Sumatra. The stages of DNA barcoding consist of DNA extraction, DNA amplification using the Polymerase Chain Reaction, electrophoresis and COI gene sequencing. The COI mtDNA gene fragment was amplified at 50oC for 45 seconds. The nucleotide length of the sample COI gene was 649 base pairs (bp). The BLASTn of BSO1 and BSO5 (Barbonymus Ogan River), and the BSG1, BSG2, BSG3 and BSG4 (Barbonymus Gerong River) showed high similarity (98.82-99.26%) with tinfoil barb (Barbonymus schwanenfeldii) from Vietnam and had a genetic distance of 0.00 (0%). The BSO2 and BSO3 from the Ogan River showed high similarity (93.59-93.66%) with the species of red tailed tinfoil (Barbonymus altus) from Cambodia and had a genetic distance of 0.03 (3%). Meanwhile, BSG5 sample from the Gerong River shows high similarity (99.56-99.85%) with silver barb (Barbonymus gonionotus) from India and Indonesia and had a genetic distance of 0.00 (0%). The phylogenetic tree construction of tinfoil barb sample taken from both rivers made three separate sub-clusters but still in the same cluster with a bootstrap value of 75%. The water quality parameter measurements include temperature ranjed from 28.8-30.0oC, water transparency 44-150 cm, salinity 0 g kg-1, dissolved oxygen 3.60-5.70 mg L-1, pH 5.6-7.8, TDS 0.02-0.04 mg L-1, ammonia 0.01-0.06 mg L-1, alkalinity 40-50 mg L-1 and current velocity 0.09-0.14 cm s-1.
