AKTIVITAS PREBIOTIK TEPUNG UBI JALAR UNGU (IPOMOEA BATATAS L.) TERHADAP LACTOBACILLUS PLANTARUM DALAM MENGHAMBAT PERTUMBUHAN ESCHERICHIA COLI
Purple sweet potato flour contains oligosaccharide compounds in the form of raffinose, which can act as natural prebiotics. The research was conducted by analyzing the effect of purple sweet potato flour before modification and after modification with fermentation and high pressure temperature on the growth of L. plantarum using Design Expert. Flour quality standardization tests were carried out based on SNI 01-3751-2009. Modifications were made to the variations TUM1, TUM2, TUM3, TUM4 with variations in fermentation time of 24 hours and 36 hours, and heating temperatures of 110oC and 121oC. Then the levels of resistant starch were determined using a UV-VIS spectrophotometer. Viability, prebiotic index and prebiotic activity tests were carried out in inhibiting the growth of E. coli using the total plate count method. The purple sweet potato flour used meets the quality standards of SNI 01-3751-2009. With normal organoleptic, fineness 99.066%, moisture content 11.386% and ash content 0.5%. The average levels of resistant starch before and after modification (TUM1) were 12.995 ± 0.404 and 40.139 ± 0.282. The results of the viability test showed that purple sweet potato flour after modification was able to increase the percentage of bacteria to 13,204%. The prebiotic index values of purple sweet potato flour before and after modification were 1.087 and 1.150. The prebiotic activity values of purple sweet potato flour before and after modification were 0.998 and 1.259. The optimum condition for modified flour was obtained at 110oC and 24 hours of fermentation, and the positive value of prebiotic activity indicated that both media were effective as prebiotic candidates and were able to reduce the growth of E. coli.
