PREPARASI DAN UJI AKTIVITAS ANTIBAKTERI SUBMIKRO PARTIKEL EKSTRAK DAUN KELOR (MORINGA OLEIFERA LAMK.) DENGAN VARIASI KONSENTRASI KALSIUM KLORIDA TERHADAP BAKTERI STAPHYLOCOCCUS AUREUS
Kelor leaf has antibacterial activity because contains quercetin which is effective against the growth of Staphylococcus aureus. This study aims to determine the variation in calcium chloride concentration as a crosslinker the submicro particles produced, to increase stability and determine its antibacterial activity submicro particles. Manufacture of submicro particles using the ionic gelation method by dispersing kelor leaf extract in a chitosan solution, then drop by drop sodium alginate and calcium chloride 20 µL; 40 µL; and 100 µL. The best formula will be determined based on the highest %EE value, then particle characterization and stability test using the heating-cooling cycle method. Kelor leaf extract contains several secondary metabolite including flavonoids, phenolics, tannins, saponins, and alkaloids. The total flavonoid content of kelor leaf extract was 31,714 mg QE/g extract. The best formula (F2) of submicro particles has %EE 84,514% with characterization of particle size 347,6 nm; PDI value 0,253; and zeta potential +21 mV. The results of the stability test F2 with a percentage reduction of 4,648%. F2 antibacterial activity against Staphylococcus aureus by disc diffusion method with inhibitory zone diameter of 22,33 ± 1,53 (strong category). It can be concluded that submicro particles of kelor leaf extract have good characterization and stability, and it has strong antibacterial activity with category ≥ 21 mm.
