OPTIMASI PRODUKSI ENZIM SELULASE DARI COLLETOTRICHUM SP. DENGAN VARIASI SUMBER KARBON DAN NITROGEN MENGGUNAKAN RESPONSE SURFACE METHODOLOGY
Fungi Colletotrchum sp. is one of the fungi that can produce enzymes cellulase. The need for cellulase enzymes continues to increase because there are many of these enzymes used in various industrial fields. However, the production of these enzymes requires quite a long time and a large cost so that the optimization of process parameters production needs to be done, one of which is by optimizing nutritional factors in the form of carbon and nitrogen sources. Response Surface Methodology (RSM) can be used to find out optimal the formula for the cultivation medium for the fungus Colletotrichum sp. in producing a production response optimal enzymes. Optimization results using Central Composite Design (CCD) on RSM showed that the use of rice straw as a carbon source and yeast extract as a nitrogen source can produse an optimum value of cellulase activity of 14,41 U/ml and the optimization medium formula recommended by RSM is by use rice straw 5,764 g/L and yeast extract 5 g/L which resulted in cellulase activity value of 12,581 U/ml and 0,069 g of biomass with a desirability value 0,787.
