OPTIMASI DAN KARAKTERISASI FORMULA SEDIAAN TRANSETOSOM KLINDAMISIN HCL DENGAN VARIASI KONSENTRASI ETANOL 96% DAN TWEEN 80
Clindamycin HCl in conventional topical formulation has a problem in its formulation. It has low bioavalilability, just around 5-8% from total concentration that can be absorbed. In addition, clindamycin HCl also difficult to penetrate into stratum corneum of the skin while Propionibacterium acnes is a bacterium that resides in the sebaceous area of the skin. To overcome this problem, it is necessary to develop delivery systems such as transethosomes. This study aims to optimize the transethosomes formula with variations of ethanol 96% and tween 80 concenctration using 22 factorial design method to obtain the optimum transethosmes formula. The optimum formula determined based on entrapment efficiency, particle size and polydispersity index. Subsequently, the optimum formula was tested for ICH Q1A(R2) standard stability test using accelerated test. The optimum formula chosen based on the system uses 20% ethanol 96% and 15% tween 80. The optimum formula characterization results were obtained entrapment efficiency of 89,401±0,118, particle size of 240,933±1,488 and polydispersity index of 0,177±0,013. The stability result using ICH Q1A(R2) standard showed that transethosomes release model followed zero order, the activation energy shelf life 22.536 days at 25℃±2℃ /60% RH±5% RH and shelf life at 5℃±3℃ in 24.572 days. Based on the study, the optimum transethosome formula showed good characterization and stability.
