Lansium domesticum Corr. is a fruit from the Meliaceae family which consists of three main groups namely duku, langsat and kokosan. This germplasm plant has market potential and high economic value, besides that this plant also has many benefits. Research on DNA barcoding using the psbA-trnH intergenic spacer gene was conducted to determine genetic variation and kinship of duku and langsat. Analysis of genetic variation was carried out by looking at mutations that occurred in aligned sequences, while phylogenetic tree reconstruction was carried out using the Mega 11 application with the Neighbor-Joining method. The results of the analysis of genetic variation obtained in the form of transversion, transition and deletion mutations, while the results of the phylogenetic tree reconstruction showed the formation of 2 clusters.
