UJI AKTIVITAS ANTIOKSIDAN EKSTRAK ETANOL DAN FRAKSI DAUN KARDIA (BELLUCIA PENTAMERA NAUDIN) PADA TIKUS JANTAN GALUR WISTAR YANG DIINDUKSI CCL4
Oxidative stress can cause degenerative diseases such as cancer, diabetes, heart disease and stroke, antioxidants are needed to overcome this. One of the Indonesian plants that has the potential as an antioxidant is cardia (Bellucia pentamera Naudin). Based on the results of the phytochemical screening, kardia leaves contain secondary metabolites which have pharmacological activity as antioxidants. The purpose of this study was to determine the characteristics of the ethanol extract and fraction of cardia leaves, the effect of extracts and fractions of cardia leaves on MDA levels using the TBARS method, catalase enzyme activity using the Mates method, macroscopic picture and histopathology of the liver of CCl4-induced rats. The research method used a completely randomized design (CRD) with 6 treatment groups and 4 replications, namely positive control (Vitamin C 10 mg/kg BW), negative control (Na-CMC 1%) and ethanol extract test group (100 mg/kg BW), fraction n-hexane (100 mg/kgBW), ethyl acetate fraction (100 mg/kgBB) and ethanol-water fraction (100 mg/kgBW). The results showed that the characterization of the ethanol extract and fraction of cardia leaves complied with the requirements of the Indonesian Ministry of Health. The MDA levels of the positive control group, negative control, ethanol extract, n-hexane fraction, ethyl acetate fraction and ethanol-water fraction were 0.74; 2.57; 1.85; 1.26; 1.34 and 1.93 nmol/mL. The activity of the catalase enzyme in the positive control, negative control, ethanol extract, n-hexane fraction, ethyl acetate fraction and ethanol-water fraction respectively were 25.67; 1.403; 18.47; 24.74; 15.64 and 8.203 U/mL. Administration of the n-hexane fraction of cardia leaves had the best antioxidant effect, because the resulting effect was equivalent to the positive control group (p>0.05). The conclusion of this study is that the n-hexane fraction of cardia leaves at a dose of 100 mg/kgBW can reduce plasma MDA levels and increase liver catalase enzyme activity as well as provide good macroscopic and histopathological features of the liver.
