UJI AKTIVITAS ENZIM AZOREDUKTASE EKSTRASELULER DARI BAKTERI AEROMONAS JANDAEI
Extracellular azoreductase enzyme activity has been tested from Aeromonas jandaei bacteria. Extract of azoreductase enzyme from A. jandaei bacteria was obtained by centrifugation of liquid culture on NB media which was incubated for 2x24 hours at 37C in an incubator shaker. The results of enzyme fractionation with the highest specific activity were used to determine the optimum conditions for the extracellular azoreductase enzyme from A. jandaei bacteria with variations in incubation time, incubation temperature, pH and substrate concentration. From the activity substrate concentration, enzyme kinetics were calculated, namely Vmax and Km. The crude extract of the azoreductase enzyme had an activity of 0.025 U/mL and a specific enzyme activity of 0.00139 U/mg. The results of the fractionation obtained the highest specific enzyme activity in fraction 3 with a saturation level of 40-60% ammonium sulfate with a specific enzyme activity value of 0.0844 U/mg. Extracellular azoreductase enzyme from A. jandaei bacteria has optimum conditions at incubation time of 5 minutes, pH 7 temperature 37ºC and substrate concentration of 70µM with enzyme activity of 0.1017 U/mL. Calculation of the enzyme kinetic data obtained a Vmax value of 0.1025 U/mg and a Km value of 0.6886 M.
