UJI AKTIVITAS ANTIDIARE FRAKSI ETIL ASETAT DAUN KETEPENG CINA (Cassia alata Linn.) PADA TIKUS PUTIH JANTAN GALUR WISTAR YANG DIIDUKSI BAKTERI Escherichia coli
Chinese ketepeng leaves contain active compounds of alkaloids, saponins, tannins, steroids, anthraquinones, and flavonoids as antibacterials. This study aims to determine the antidiarrheal activity of the ethyl acetate fraction of Chinese ketepeng leaves induced by Escherichia coli bacteria. This research was conducted by experimental method. The treatment group was divided into seven groups: normal, negative control (Na-CMC), positive control (Gentamycin), and test group 1 (40), test 2 (80), test 3 (160) and test 4 (320) mg/ kg BB. The test parameters carried out in this study were stool weight, stool consistency, frequency of diarrhea, body weight, number of Escherichia coli bacteria colonies in rat feces. Ketepeng cina leaf extraction was carried out by maceration method using 70% ethanol then fractionated with n-hexane and ethyl acetate. Characterization of the ethanol extract of Chinese ketepeng leaves obtained water soluble compound content of 41.66%, ethanol soluble extract content of 81.66%, water content of 9%, drying shrinkage of 8.71%, total ash content of 6.33%, acid insoluble ash content 3.8230% , 75 cfu/g microbial contamination test, Pb metal contamination test < 0.068 mg/kg. The parameter of acid insoluble ash content obtained was 3.823% which did not meet the specified requirements (≤0.7%). The total tannin content was 6.315 mg/gCE and the total flavonoid content was 4.21 mgQE/g. The results showed that Chinese ketepeng leaves had antidiarrheal activity where doses were 40 (30.23%), doses 80 (36.10%), doses 160 (42.28%) and doses 320 (52.87%). This showed that the antidiarrheal effect of the ethyl acetate fraction of Chinese ketepeng leaves at a dose of 320 mg/kg BW was not significantly different from the positive control drug gentamicin (P>0.05). The number of E. coli colonies when diarrhea cured for doses of 40, 80, 160, and 320 mg/kg BW were 111.2 x 105 (cfu/g) respectively; 110.2 x 105 (cfu/g); 107.4 x 105 (cfu/g); and 104.4 x 105 (cfu/g). The effective dose (ED50) of the ethyl acetate fraction of Chinese ketepeng leaves as an anti-diarrhea is 274.4187 mg/kgBB.
