UJI TOKSISITAS AKUT FRAKSI ETIL ASETAT BUAH LAKUM AIR (Ludwigia octovalvis (Jacq.) P.H. Raven) PADA TIKUS PUTIH JANTAN GALUR WISTAR DENGAN METODE FIXED DOSE PROCEDURE
Ethyl acetate fraction from lakum air (Ludwigia octovalvis (Jacq.) P.H. Raven) fruit has α-glucosidase inhibitory activity as antidiabetic. The results of phytochemical screening from ethyl acetate fraction of lakum air fruit contain flavonoids, saponins, phenolics, and tannins. Based on the useful compounds, we need to know their safety, supported by toxicity testing. In this research, an acute toxicity test was done of ethyl acetate fraction from lakum air fruit on Wistar strain male white rats using a fixed dose procedure method. The preliminary test results determined the main test dose at 2000 mg/kgBW. The results of the main test observations showed that the normal group and the test dose group at 2000 mg/KgBW did not show any toxic symptoms or death of the test animals. The toxic dose range of ethyl acetate fraction from lakum air fruit is more than 2000 mg/KgBW which is practically non-toxic. Ethyl acetate fraction from lakum air fruit at 2000 mg/KgBW did not affect the macroscopic organs of the kidney and heart (p>0.05) but had a significant effect on the shape and weight of the liver (p0.05). The levels in the normal group SGOT 179.94±33.13 U/L, SGPT 61.9±7.90 U/L, creatinine 0.742±0 0.05 mg/dL, and urea 37.14±8.50 mg/dL. While the test dose group at 2000 mg/KgBW SGOT 135,076±50,40 U/L, SGPT 66,698±13,53 U/L, creatinine 0,646±0.04 mg/dL, and urea 29,14±0.94 mg/dl. The ethyl acetate fraction of lakum air fruit at 2000 mg/KgBW did not show the effect of microscopic damage to the pancreas.
