Pain is a sign that something wrong in the body system such as tissue problem, also it is one of the most complaints when patient sees doctor. Pain can be treated with analgesic drug. One of plant which is widely used as an herbal medicine is Peronema canescens. The leaves of P. canescens has been reported contained betulinic acid, glycoside, β-sitosterol, stigmaterol and phytol. The aims of this research was determined analgesic effect of P. canescens leaves. The leaf was extracted by maceration method using ethanol 96% solvent. The analgesic effect was determined by hot plate and formalin method with latency time and licking time as a pain response parameter. The result on the characterization of extract ethanol P. canescens leaf had qulified the requirement. This research showed that ethanol extract of P. canescens leaves at the dose 100, 200 and 400 mg/KgBW has percentage of analgesic 35.28%, 44.23% and 45.16% respectively. Dose 200 and 400 mg/KgBW were better than diclofenac sodium as positive control (percentage of analgesic was 39.93%). Extract ethanol P. canescens leaf can reduced pain only on the second phase of formalin test. Percentage of analgesic with formalin test on the first phase and second phase respectively were 6.25% and 57.79%. Mechanism action of analgesic from extract ethanol P. canescens leaf influenced by dopamin receptor.
