UJI AKTIVITAS ANTIDIABETES FRAKSI ETIL ASETAT BUAH LAKUM AIR (Ludwigia octovalvis (Jacq.) P.H. Raven) TERHADAP TIKUS PUTIH JANTAN GALUR WISTAR TERINDUKSI STREPTOZOTOSIN
The research on the antidiabetic activity of the ethyl acetate fraction from lakum air (Ludwigia octovalvis (Jacq.) P.H. Raven) fruit in streptozotocin-induced male white rats of wistar strain was aimed to determining the decrease in blood glucose levels, ED50 (Effective Dose 50) and the histopathological effect on the pancreas from diabetic rats type 2 treated with the ethyl acetate fraction of lakum air fruit. The group test animals used were divided into 7 groups consisting of a normal group that was only given 0,5% Na CMC suspension without experiencing diabetes, a positive control group using acarbose dose of 4,94 mg/kgBW as a comparison, a negative control group using 0,5% Na CMC suspension and the ethyl acetate fraction of lakum air fruit with various doses of 20, 40, 60, and 80 mg/kgBW. The results showed that the AUC0-15 value and the percentage decrease in blood glucose levels for the positive control were 1429,20 and 70,16%, the ethyl acetate fraction treatment group with doses of 20, 40, 60, and 80 mg/kgBW respectively 3849,30 and 19,62 %; 2838,60 and 40,72%; 2321,40 and 51,52%; 1656,00 and 65,42%. The ethyl acetate fraction of lakum air fruit with the best antidiabetic activity was the highest dose (80 mg/kgBW). Analysis of the decrease in blood glucose levels in the 40, 60, and 80 mg/kgBW did not show a significant difference compared to acarbose (p>0,05). The ED50 of ethyl acetate fraction from lakum air fruit was 57,67 mg/kgBW. Observation of 80 mg/KgBW rats pancreatic cell necrosis score was as good as the positive group.
