One of the plants that has the potential as a natural antioxidant is balik angin plant (Mallotus paniculatus (Lam.) Mull. Arg.). Based on the results of the phytochemical screening, the leaves of balik angin contain flavonoids, steroids, triterpenoids, saponins, tannins and phenolics. This study was aimed to determine the characteristics of the extract and the antioxidant activity of the ethanol extract and leaf fraction of balik angin with parameters of MDA levels using the TBARS method, catalase enzyme activity using the Mates method, macroscopic and histopathological features of the livers of CCl4-induced Wistar male rats. The research method used a completely randomized design (CRD) with male white rats Wistar strain which were divided into 6 treatment groups, namely positive control (10 mg/kgBW vitamin C), negative control (1% Na CMC), ethanol extract, n- hexane, ethyl acetate fraction and ethanol-water fraction at a dose of 300 mg/kgBW. The results of the characterization of the ethanol extract met the requirements of the Ministry of Health of the Republic of Indonesia. The results showed that MDA levels of the positive control, negative control, ethanol extract, n-hexane fraction, ethyl acetate fraction and ethanol-water fraction were 0.740; 2,579; 1.109; 1.144; 0.947 and 1.109 nmol/mL. The activity of catalase enzyme in the positive control, negative control, ethanol extract, n-hexane fraction, ethyl acetate fraction and ethanol-water fraction respectively were 25.675; 1.403; 19.073; 15,649; 24.676 and 8.963 U/mL. The macroscopic and histopathological appearance of the liver showed an improvement after administration of the extract and fraction of the leaves of balik angin. The ethyl acetate fraction showed the best antioxidant potential and the resulting effect was equivalent to the positive control group of vitamin C (p>0.05).
