Antioxidants have a function to prevent the occurrence of oxidation processes caused by oxidation free radicals by releasing hydrogen atoms or electrons. Therefore, it is necessary to conduct further research on antioxidant-producing plants. Jambu nasi-nasi (Syzygium zeylanicum (L.) DC) has potential as an antioxidant producer because it contains compounds such as flavonoids, terpenoids, tannins, phenols, polyphenols, and steroids. The purpose of this study was to determine the antioxidant activity of fractions, compound classes, and IC50. This research was conducted from August to December 2022 at the Laboratory of the Biology Department, FMIPA, Sriwijaya University. The research methods used were extraction, fractionation, antioxidant activity test of the fractions using the thin layer chromatography method, compound isolation, antioxidant activity test of the fractions using a UV-Vis Spectrophotometer, and determination of the IC50 value using the DPPH method. The results of this study are presented in the form of tables, figures and graphs. The results of thin layer chromatography are presented in the form of chromatogram images and linear regression analysis are presented in the form of linear regression graphs. Jambu nasi-nasi leaves (Syzygium zeylanicum (L.) DC) have strong antioxidant activity in the n-hexane and also ethyl acetate fractions as indicated by the presence of dark yellow spots on the chromatogram of the fraction and the IC50 value. Pure isolates suspected to belong to the class of terpenoid compounds are N1, N2.1, N2.3, N3, E3.1, and E3.4. Isolate N6 belongs to the flavonoids, E2.6 belongs to the tannins. Very strong antioxidant activity was found in isolates N1, N2.1, N6, E3.1, E3.4, and E4, with IC50 values of 5.27 ppm, 13.74 ppm, 16.91 ppm, IC50 43, respectively 68 ppm, and 26.08 ppm. Strong antioxidant activity was found in isolates N2.3 and E2.6 with IC50 values of 53.07 ppm and 81.49 ppm. N3 IC50 value of 196.68 ppm, weak antioxidant.
