The stages of developing traditional medicines into phytopharmaca include the selection stage, preclinical testing, simple standardization, identification determination and standardized preparation preparation as well as clinical trials. Purple sweet potato leaves contain flavonoids and tannins, flavonoids which are antioxidants which function to inhibit cholesterol synthesis. This study aims to determine the acute toxicity of the ethanol fraction of purple sweet potato leaves (Ipomoea batatas L.) in vivo in male Wistar rats using the fixed dose procedure method. In this study, 2 tests were carried out, namely the preliminary test and the main test (2 groups, normal and treatment at a dose of 2000 mg/KgBB). The toxic dose range of the ethanol fraction of purple sweet potato leaves that causes acute toxicity is >2000 mg/KgBW so that it is included in the practically non-toxic category. Based on the research results in the main test showed that there was no death or toxic symptoms in the two groups of test animals. The average levels of normal biochemical parameters were SGOT 173.94 ± 4.327 U/L, SGPT 65.78=15.54 U/L, creatinine 0.65 ± 0.015 mg/dl and urea 33.42 ± 3.061 mg/dl. The average test dose group was 2000 mg/Kg BW for SGOT 171.14 ± 25.98 U/L , SGPT 96.36 ± 32.42 U/L , creatinine 0.63 ± 0.03 mg/dl, and urea 39 .94 ± 6.60 mg/dl . Administration of the ethanol fraction of purple sweet potato leaves at a dose of 2000 mg/KgBW had no significant effect on SGOT, SGPT, creatinine and urea (p>0.05). Administration of the ethanol fraction of purple sweet potato leaves at a dose of 2000 mg/KgBW did not show any effect on macroscopic, microscopic and histopathological damage to the liver, kidney and heart.
