IDENTIFIKASI MOLEKULER Citrus SP. (RUTACEAE) KOLEKSI KEBUN RAYA BOGOR BERDASARKAN SEKUEN DNA BARCODE rbcL DAN psbA-trnH INTERGENIC SPACER
Molecular identification is identification with a molecular approach, one of which is with a DNA sequence called a DNA barcode. DNA barcode is short sequence of DNA that have been standardized and can be used for identification. Molecular identification is generally done to confirm the identity and classification of existing species as well as for species that are not yet identified. Citrus sp. that Bogor Botanical Garden collected from West Sumatra has not been identified to the level of the species because it has not produced generative organs. Hence, the identification of Citrus sp. molecularly is more effective to do. This study aims to compare the primary discrimination ability of rbcL and trnH-psbA (IGS) against Citrus sp. collection of Bogor Botanical Garden from West Sumatra as well as identifying species-level taxons from Citrus sp. Bogor Botanical Garden collection from West Sumatra. This research was carried out in July 2022 – Februari 2023, at the Treub Laboratory, Plant Conservation Research Center, Botanical Garden and Forestry – National Research and Innovation Agency, Jl. Ir. H. Juanda No. 13 Bogor, West Java, Indonesia. The research methods are DNA isolation, electophoresis, DNA amplification, DNA sequencing, data analysis and data presentation. This study used primer rbcL and psbA-trnH intergenic spacers. The results of the study obtained that rbcL primers have not been able to discriminate against Citrus sp. at the species level whereas primary psbA-trnH primers classifies Citrus sp. to Citrus japonica based on the philogeny tree and leaf morphology of Citrus sp.
