ISOLASI SENYAWA TURUNAN POLIKETIDA DARI FRAKSI ETIL ASETAT BATANG TUMBUHAN TITHONIA DIVERSIFOLIA
Tithonia diversifolia is a plant from the Asteraceae family of the genus Tithonia, originating from North and Central America. T. diversifolia has been reported as a biofumigant, anti-TMV (Tobacco mosaic virus), insecticide, antidiabetic, and antioxidant both from extracts, fractions, and metabolites that were successfully isolated. The content of secondary metabolites from T. diversifolia leaves and aerial parts was reported more than the stem. Based on this knowledge, the research will be carried out to isolate and identify the secondary metabolites present in the stem of this plant. The extraction process was carried out by the maceration method, using methanol as a solvent. Then proceed with the liquids fractionation process using n-hexane solvent followed by ethyl acetate solvent. The process of isolation or separation and purification of compounds was carried out using VLC (vacum liquid chromatography) and GCC (gravity chromatography column). The isolated compound obtained as a white crystals (38.8 mg) and has a melting point of 147.3℃-148.9℃. Structural characterization of the isolated compound was concluded using UV-Vis spectroscopy, IR, and NMR spectroscopy (1H-NMR, 13C-NMR, HSQC, and HMBC). Based on the spectroscopic data and comparison with literature data, concluded that the isolated compound was a polyketide derivative, namely De-O-methylacetovanillochromene.
