UJI TOKSISITAS DAN ISOLASI SENYAWA STEROID DARI FRAKSI N-HEKSANA BATANG TUMBUHAN Passiflora foetida
Passiflora foetida is a vine plant belonging to Passiflora genus that reported to have biological activity as an antibacterial, anti-ulcer, antidiabetic, antioxidant and anti-inflammatory. Studies on steroid compounds and toxicity assays have never been reported from P. foetida stems. Therefore, this study was conducted to isolate steroid compounds from n-hexane fraction and toxicity assays from methanol extract, n-hexane fraction, ethyl acetate fraction and the isolate compounds using in vivo method of Brine Shrimp Lethality Test (BSLT). Extraction by maceration using methanol solvent and partitioned by fractionation using n-hexane and ethyl acetate solvents. Separation and purification of the compounds from n-hexane fraction were performed using vacuum liquid chromatography and gravity column chromatography. The result of the separation were 2 isolate compounds in the form white crystalline, namely isolate D (76.8 mg) and isolate G (9.1 mg). Based on IR, GC-MS and NMR spectroscopy analysis showed that both isolates were steroid compounds. The toxicity assay for the methanol extract, n-hexane fraction and ethyl acetate fraction gave LC50 values respectively 361.92, 72.71 and 38.13 ppm showed that the methanol extract had a medium toxicity level, while the n-hexane and ethyl acetate fractions showed a very toxicity level. The toxicity assay for the isolate G gave LC50 value of 21.58 ppm showed more active toxicity than isolate D with LC50 value of 121.22 ppm.
