AKTIVITAS ANTIOKSIDAN DARI FRAKSI ETIL ASETAT RANTING TUMBUHAN RUKAM (FLACOURTIA RUKAM) DAN IDENTIFIKASI SENYAWA PADA FRAKSI AKTIF MENGGUNAKAN GC-MS
Flacourtia rukam is one of the traditional medicinal plants used in the treatment of various diseases. The disease can be caused by various factors, one of which is due to free radicals. Antioxidant compounds are scientifically known to be able to inhibit free radicals. This research was conducted to test the antioxidant activity of the ethyl acetate fraction of F. Rukam twigs. The extraction using maceration method with solvents of step gradient polarity (n-hexane and ethyl acetate). Separation and purification were then carried out on the ethyl acetate extract using gravity column chromatography and continued with antioxidant activity tests using the DPPH (1,1 diphenyl-2-picrylhidazyl) method. Separation of the ethyl acetate extract obtained 5 fractions with the highest antioxidant activity in the F5 fraction (IC50 47.81 mg/L). Further separation and purification were carried out on the F5 fraction to obtained 5 subfractions and the strongest antioxidant activity test results were in the F5.3 subfraction (IC50 43.36 mg/L). Identification of compounds from the F5.3 fraction (33.1 mg) which are active antioxidants was carried out using GC-MS spectrophotometry. Analysis by GC-MS obtained compounds in the form of unsaturated aliphatic hydrocarbons namely 9-nonadecene and 1-octadecene with Similarity Index (SI) 905 and 902. Aliphatic hydrocarbon compounds are classified as hormones in plants so it is assumed that compounds that have antioxidant activity are not identified.
