ISOLASI SENYAWA BIOAKTIF DARI JAMUR ENDOFITIK DAUN SUNGKAI (Peronema canescens Jack.)
Sungkai leaves (Peronema canescens Jack.) has been used by the community for a long time as a traditional medicine for fever, stomachache, mouth antiseptic, and toothache medicine. This research was conducted to isolate and identify bioactive compounds from endophytic fungi of sungkai leaves. The research was started by isolating and purifying endophytic fungi from fresh sungkai leaves, then identified morphologically. Then the endophytic fungi were cultivated for 4 weeks on PDB media and extracted to obtain a concentrated extract of ethyl acetate. The extract obtained will be tested for antioxidants using the DPPH method. Extracts with the highest antioxidant potential will be isolated by chromatographic technique and the endophytic fungi will be identified molecularly. The pure compound obtained will be determined spectroscopically for its structure.The results of the isolation and purification of endophytic fungi obtained five isolates of endophytic fungi with different species. The five fungal isolates were cultivated and extracted to obtain a concentrated extract of ethyl acetate. The ethyl acetate extract was tested for antioxidant activity and it was found that the SD3 isolate had the highest activity (IC50 = 22.32 μg/mL). Based on the phylogenetic tree construction, the SD3 endophytic fungus is Colletotrichum gloeosporioides. The results of isolation of the SD3 extract using column chromatography and analysis using thin layer chromatography obtained pure compounds in the form of compounds with an aromatic group that has a para-substituted benzene ring, ester and hydroxyl groups in the form of a white solid (74 mg) which has strong antioxidant activity with IC50 = 20. 87 μg/mL.
