EFEK NEFROTOKSIK GENTAMISIN TERHADAP TIKUS WISTAR.
Gentamicin is one example of an aminoglycoside that gives its users nephrotoxic side effects. Although gentamicin has had nephrotoxic effects until now there is still little research on the effects of gentamicin on the kidneys and making gentamicin as an induction drug to make animals try models of renal fibrosis. The purpose of this study was to determine the nephrotoxic effect that leads to renal fibrosis due to the induction of high doses of gentamicin against Wistar rats. The experimental study, in vivo, has carried out in the Animal House laboratory and the Medical Laboratory Center Palembang from September–October 2022. There are 4 treatment groups namely placebo (NaCl 0,9%), 80 mg/kgBW of KIG I, 120 mg/kgBW of KIG II, and 240 mg/kgBW of KIG III single dose intraperitoneally of 8 wistar rats in each group. All Wistar mice of each group were taken blood samples on days 0,3,7, and 14. The results of the study were analyzed using the IBM SPSS Statistics 25 application. The results of the study using the Levene test obtained the body weight of homogeneous rats (p>0.05). The nephrotoxic effect of gentamicin showed that KIG I and KIG II were significant in raising the average creatinine and urea level on day 0 and day 14 of treatment (p50% of experimental animals in KIG III after treatment and already reached Lethal Dose 50 (LD50). In conclusion that KIG I and KIG II have a meaningful nephrotoxic effect in raising creatinine and urea levels leading to renal fibrosis.
