DNA BARCODE IKAN SELINCAH (Belontia sp.) BERDASARKAN GEN SITOKROM C OKSIDASE SUBUNIT I (SOI).
The combtail fish (Belontia sp.) is a type of fish that lives in groups in river or swamp waters. The combtail fish are included in the Belontiidae family. This family is presented only by genus Belontia and two species Belontia hasselti and Belontia signata. The purpose in this research was to obtain the Cytochrome C Oxidase (COI) gene sequences, the percentage of nucleotide identities of coumbtail with genetic data on GenBank, genetic distances, and phylogenetic trees between combtail species, and to know the chemical-physical characteristics of the water in the fish habitat in Belida River, Gelumbang District, Muara Enim and the swamp of Tanjung Baru Village, North Indralaya District, Ogan Ilir. The research has been carried out in January 2022. The COI gene was amplified by PCR with optimum annealing temperature 52oC for 1 minute and 35 cycles. Sequence length of the COI gene of Belontia sp. was 672 base pairs (bp) nucleotide. Based on BLASTn analysis compared to GenBank database, combtail sample from Belida River and swamp of Tanjung Baru Village had the highest similarity of 95.23% and genetic distance of 0.052 with Belontia hasselti from Indonesia. Physical chemical of water characteristics of the combtail were temperature 28.3-31.8oC, water transparency 23-36 cm, dissolved oxygen 4.3-5.5 mg L-1, pH 5-6 and total alkalinity 20-28 mg L-1 CaCO3.
