Background: Down syndrome is the most common chromosomal abnormality in people with intellectual disabilities. A lot of stigma in society can occur due to inadequate knowledge and negative attitudes that a prospective doctor has. This study aims to identify the knowledge and attitudes of medical students in Indonesia towards Down Syndrome. Methods: This study is a descriptive observational study with a cross-sectional design. The samples are medical students in Indonesia that apply inclusion criteria by taking samples using the cluster sampling technique. The instrument in this study is the Knowledge and Attitude towards Down Syndrome Questionnaire by Alhaddad et al. Results: The results showed that students had adequate knowledge (98.7%) and positive attitude (93.5%). Percentage of adequate knowledge and positive attitude at the age of ≤20 years (99.6%) and (94.9%), >20 years (98.2%) and (92.7%). Then in women (98.8%) and (93.8%), in men (98.6%) and (92.6%). Then pre-clinical (98.9%) and (93.9%), clinical (97.5%) and (91.1%). From public universities (99.5%) and (94.6%), private (97.3%) and (91.5%). Excellent accreditation (99.1%) and (94.1%), very good (97.7%) and (92.4%), good (100%) and (92,1%). Region one (98.6%) and (94.4%), region two (99.3%) and (89.9%), region three (99.2%) and (92.9%), region four (98.1%) and (96.2%). With family history (95.6%) and (88.9%), without a family history (99%) and (93.9%). With a history of interaction (98.3%) and (92.7%), without a history of interaction (99.1%) and (94.3%). Conclusion: Medical students in Indonesia have adequate knowledge and positive attitude towards Down Syndrome. Keywords: Down Syndrome, Knowledge, Attitude, Stigma
